Cellular localization of herpes simplex virus 1 UL24 protein in the absence of viral infection. The localization of UL24 fused to a haemagglutinin epitope tag (HA-UL24) in transiently transfected COS-7 cells was assessed by confocal microscopy. Nuclei were stained with Draq5 (blue) and HA-UL24 was visualized by indirect immunofluorescence using a primary antibody specific for HA and a secondary antibody conjugated to the fluorophore Alexa-488 (green). Top, negative control showing staining with the secondary antibody only; middle, the major staining pattern observed for HA-UL24; bottom, the minor HA-UL24 staining pattern.
